WRIB 2022: Optimization and standardization of RNA-seq operation to get high-quality sequencing data with heavily degraded RNA samples from tumor biopsies

We standardized an RNA-seq operation method under Good Lab Practice (GLP) standards, which has been consistently able to get high-quality data, particularly for degraded RNA (1-10ng total RNA). Here, we presented three key skills to control the operation process, including the total RNA quantification, library kit selection and data analysis process. In general, this standardized method has been working very well with all the samples we have tested so far, even though only tiny total RNA contents were obtained to start with.
